How can I tell if my art niche is big enough to build a real business?

One heuristic: Go to a bookstore magazine section and see if there are multiple magazines devoted to your topic—that indicates a large market. Another approach: Check if other brands and manufacturers already target that audience. For example, college alumni are highly targetable because companies sell sweatshirts, hats, posters, and more to them. If other businesses are successfully marketing to an audience, it’s a good sign the market exists and is reachable.
